TG1358_mpz-EGFP_transgene map To produce mpz promoter-EGFP transgenic (mpz-EGFP) medaka, we amplified the 2.9-kb upstream regulatory region of the medaka mpz gene using the mpz promoter Sal I forward primer 5′- GTCGACTGGCTTCCATTCTTGACTCC-3′ and mpz promoter Kpn I reverse primer 5′-GGTACCGGTTGTTCCTGGCTTCCCAA-3′. The amplified fragment was cloned into a TA cloning vector (pCR-TOPO Vector, Thermofisher). We then digested the fragment with Sal I and Kpn I, and subcloned it into the Sal I and Kpn I sites of the I-SceI backbone vector, in which EGFP Express/SV40 polyA (Clontech) was inserted into the Hind III and AflII sites of pBSKI2 (Amagen). The Ensemble Genome database indicated the mpz gene in Japanese medaka fish HdrR. |
