FLFII-Tg(actb-EGFP)19

Strain information
Strain ID TG1337
Strain Name FLFII-Tg(actb-EGFP)19
Strain Type Transgenics
Characteristics Single transgene integration on chromosome 19, strong ubiquitous EGFP fluorescence. Paternal expression in blastula visible. Maternal expression visible until 6 dpf (Iwamatsu stage 36) also in non-transgenic offspring. see: Froschauer, A., Sprott, D., Gerwien, F. et al. Effective generation of transgenic reporter and gene trap lines of the medaka (Oryzias latipes) using the Ac/Ds transposon system. Transgenic Res 21, 149.162 (2012). https://doi.org/10.1007/s11248-011-9514-x
Transgene: minDS-Sites of Ac/Ds transposon vector pMDS6 flanking the actb-promoter (including intron1) and actb-UTR of medaka driving EGFP of Clontech. For pMDS6 see: Emelyanov et al (2006) Trans-Kingdom transposition of the maize dissociation element. Genetics 174(3):1095.1104
Notes Please note that we will use OK-Cab (MT830) for artificially insemination with frozen sperm of this strain.

TG1337_actb-EGFP
Establishment of actb-EGFP transgenic medaka. a,b Embryos at 6 dpf (stage 36). Embryos were injected with plasmid DNA alone (a) or with plasmid DNA and transposase mRNA (b) at the 1.2-cell-stage. EGFP-expressing individuals were raised and mated to non-transgenic fish. c Zygotic transgene expression in an F1 embryo of a male founder begins at early blastula stage. d At hatching stage, the strongest expression is seen in the central nervous system and the digestive tract.
