2008年4月1日より枯草菌遺伝子破壊株の分譲を開始しました。この遺伝子破壊株ライブラリーは、Kobayashi et al.(2003 PNAS vol.100 4678-4683)で新規に報告された遺伝子破壊株をもとに作製されたものです（野生株1株、遺伝子破壊株2514株）。本プロジェクトでは、遺伝子破壊株 2514株について遺伝子破壊確認のPCR実験を行い、確認できた株を分譲します。 遺伝子破壊確認のPCR実験は以下のように行いました。一次検定として、すべての株について、破壊された遺伝子の開始コドンとpMUTINベクターで挟まれた領域（１）と破壊された遺伝子の終止コドンとpMUTINベクターで挟まれた領域（２）の二つの領域についてプライマーを作製してPCR実験を行い、増幅の確認できた株を分譲可としました。 次にこれらの領域のどちらでも増幅しなかった株について、二次検定として、破壊された遺伝子の開始コドンと終止コドンで挟まれた領域（pMUTINベクター配列を含む）（３）でPCR実験を行い、遺伝子とpMUTINベクター配列をあわせた10kb程のDNA断片の増幅が確認できた株を分譲可としました。 以上の二つの検定により、寄託された2515株のうち、野生株1株及び、遺伝子破壊が確認できた2093株について分譲を行います。（青木敬太） |
